But yes - as legato said, curios are gotten via both a vendor and various challenges can be unlocked, progression in the story provides more flexibility and difficulty.
Progression also unlocks more challenges you can do in town as well.
- There are curios to collect (you can choose a single curio to provide certain effects on your run).
- You unlock more rune slots, allowing you to make more diverse/stronger decks or equip stuff like immunities.
- Beating a dungeon means other dungeons get harder.
From the top of my head, that's the main stuff, but honestly, the cards are plenty too.
