Haven't you tried this yet? Well, this developer's hint is pinned on the forums:

I bet you tried this already and had no luck, eh? Well, I have a little trick for you.

  • Change Compatibility Mode

Check the game folder again (probably at C:\Program Files (x86)\Steam\SteamApps\common\Deadly Premonition The Director's Cut) and set both DP.exe and DPLauncher.exe to run in compatibility mode for: Windows XP (Service Pack 3). Now try running the game again and voilà: it is finally running smoothly as it should!

Remember this fix worked on Win10. Give me some feedback to confirm whether this is working or not. Good luck!

i'm using DPfix as well as the 4GB patcher and the game was crashing at a specific point around the end of the hospital section. Running in this compatibility mode has helped a lot so far.

Changing compatibility mode isn't a good advice, especially when the game runs without issues on Windows 10.

Hey Llama,
The only scenario where you get this message is when you are running DP.exe itself. Try setting both files to run as WinXP SP3 and run the game through Steam.
