On March 15, 44 BC, a group of senators summoned Caesar to the forum to read a draft petition asking him to return power to the Senate. But the petition was a hoax. Marcus Antony, who had partially learned of the assassination plot the night before from a terrified Liberator, Servilius Casca, and fearing things would get worse, went to the forum to intercept Caesar on the steps. But Caesar's path was blocked by a group of senators as he passed through the Pompeian Theater on the Campus Martius, and Caesar was led into a room adjacent to the east portico.

As Caesar began to read the false petition, Tillius Cimber, who had presented it to him, pulled down Caesar's toga. As Caesar shouted at Cimber, "But this is an atrocity!" ("Ista quidem vis est!"), Casca drew his dagger and slashed the dictator's throat from side to side. Caesar immediately turned around and grabbed Casca's arm, saying, "Casca, you traitor, what are you doing?"[65] Casca, frozen with fear, shouted in Greek, "My brothers, help me" ("ἀδελφέ, βοήθει!", "adelphe, boethei!"). Just then the rest of the group, including Brutus, began to stab Caesar. Caesar tried to flee but, blinded by blood, tripped and fell to the ground; the men kept stabbing him with their daggers until Caesar was defenseless on the lower steps of the portico. According to Eutropius, there were more than sixty men involved in the assassination. Caesar was stabbed 23 times.[66] According to Suetonius, a physician proved that only the second of his wounds, a wound to the neck, was fatal.[67]

It is not known for certain what the dictator's last words were and this has been a matter of debate among scholars and historians throughout history. The most commonly known version is the Latin phrase Et tu, Brute? ("You too, Brutus?"), taken from Shakespeare's play Julius Caesar. In fact, exactly as the first part of a macaronic line: "Et tu, Brute? so fall down (die) Caesar." Shakespeare's version is followed by that of the Roman historian Suetonius, who claimed that Caesar's last words were the Greek phrase "καὶ σύ, τέκνον;"[68] (pronounced "Kai su, teknon?": in Turkish "You too, my child? ").[69] On the other hand, Plutarch reports that Caesar said nothing and that when he saw Brutus among the assassins he pulled his toga over his head.[70]

Plutarch claims that after the assassination, Brutus wanted to go in front of his fellow senators to say something, but they preferred to flee the building.[71] Brutus and his entourage marched shouting towards the Capitol Hill and called out to their beloved city: "People of Rome, we are free once more!". These cries of joy were met with silence as the citizens of Rome, already realizing what was about to happen, locked themselves in their homes.
